fa بیان ژنPPARγ در سلول‌های چربی تمایز یافته از سلول‌های بنیادی مزانشیمی مغز استخوان انسانی مدیریت خدمات بهداشتی درمانی Hospital Managment پژوهشی اصيل Original Research <p align="justify"><strong>زمینه و هدف: </strong>اخیراً، چاقی به عنوان عامل خطر تهدید کننده برای بیماری‌های شناخته شده‌ای از جمله بیماری‌های قلبی _ عروقی، دیابت غیر وابسته به انسولین و سرطان‌های شایع به حساب می‌آید. بنابراین درک فرایندهای تمایز ادیپوست و بیان ژن های دخیل در این روند حیاتی است. </p><p align="justify"><strong>روش بررسی </strong><strong>:<i> </i></strong>در این مطالعه، سلول‌های بنیادی مزانشیمی مغز استخوان انسانی( H-MSCs ) به وسیله شیب غلظتی فایکول جدا شده، مارکرهای سطحی این سلول‌ها به وسیله فلوسیتومتری تایید گردیده و تمایز به چربی و استخوان به وسیله دستورالعمل دگزامتازون انجام گرفت و به وسیله رنگ آمیزی تایید شد. سپس بیان کمی و کیفی ژن γ PPAR به عنوان مهمترین عامل نسخه برداری در تمایز چربی قبل و بعد از تمایز به ادیپوسیت به وسیله RT-PCR و qPCR بررسی و تحلیل آماری به وسیله paired t test و با کمک نرم افزارهای Pfaffle و graph pad انجام شد. </p><p align="justify"><strong>یافته‌ها: </strong><strong></strong>بیان این ژن بعد از تمایز سلول‌های بنیادی مزانشیمی مغز استخوان انسانی به سلول چربی افزایش قابل ملاحظه‌ای را نشان داد(0/05>p ). </p><p align="justify"><strong>نتیجه‌گیری: </strong><strong></strong>ژن PPARγ به عنوان یکی از عوامل با اهمیت در تمایز سلول‌های MSCs به آدیپوسیت عمل می‌کند و مهار بیان این ژن برای جلوگیری از چاقی به عنوان یک ایده برای درمان در آینده پیشنهاد می‌شود. </p> <p align="justify"><strong>Background and Aim: </strong>Obesity is now considered as one of the main risk factors of certain known diseases such as cardio-vascular diseases, non- insulin-dependent diabetes, and common cancers. Moreover, the increase of white fat tissue is known as a main factor in the obesity process, in terms of physiology and pathology. Therefore, the understanding of adipocytes differentiation processes is crucial. <strong></strong></p><p align="justify"><strong>Materials<i> </i>and<i> </i>Methods: </strong><i>In this study, mesenchymal stem cells (MSCs) were isolated from human bone marrow by ficol-gradient, and then, their surface markers were confirmed by flow cytometry. Osteoblastic and adipocytes differentiation were done by dexamethasone protocol and confirmed by staining. Then qualitative and quantitative expressions of PPARgamma (PPAR-γ) gene as an important transcription factor in the differentiation of fat were studied by RT-PCR and REAL TIME PCR before and after differentiation into adipocytes. For statistical analysis, paired t-test was performed, using pfaffl and graph pad software. </i></p><p align="justify"><strong>Results: </strong><strong></strong>PPAR-gamma gene expression showed a significant increase after differentiation of human bone marrow mesenchymal stem cells into adipocytes (p<0/05). </p><p align="justify"><strong>Conclusion: </strong>According to the results, the PPAR-γ gene acts as one of the important factors in the differentiation of MSCs into adipocytes.
